Molecular Glues

Molecular glues are small molecules that promote or stabilize interactions between proteins that would otherwise not associate. By enhancing the binding between a target protein and an E3 ubiquitin ligase, molecular glues induce ubiquitination and proteasomal degradation of the target. Unlike bifunctional degraders, molecular glues rely on conformational stabilization rather than a linker to bring proteins together.

Molecular glues represent an emerging class of targeted protein degraders with relatively simple structures and favorable drug-like properties. Their ability to modulate protein–protein interactions opens new opportunities for targeting challenging disease-associated proteins. Ongoing discoveries in chemical biology are rapidly expanding the scope of molecular glue therapeutics across oncology and other disease areas.
