This is the reversible-terminator chemistry underlying cyclic sequencing-by-synthesis: 3'-O-azidomethyl-dNTPs are still recognized by DNA polymerase as substrates and act as reversible terminators, so incorporation adds one base and stops extension. After each incorporation and readout, the azidomethyl cap is removed with a water-soluble phosphine (TCEP) to regenerate the 3'-OH so the polymerase reinitiates.
