How do molecular glues differ from PROTACs?
Molecular glues are single small molecules that stabilize a protein-protein interaction, often between an E3 ligase and a neosubstrate. PROTACs are bifunctional molecules with a target ligand and E3 ligand connected by a linker.
What is a neosubstrate?
A neosubstrate is a protein recruited to an E3 ligase in the presence of a molecular glue, leading to ubiquitination and degradation in the relevant biological system.
